Differentiation of Human Cytomegalovirus Strains by Using PCR and RFLP

Abstract
|
|
|
A direct typing method was developed for human cytomegalovirus (hCMV) by analysis of polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) of the alpha sequence, known to be a cleavage/packing cis-functional
sequence.
The
alpha sequence was directly amplified by PCR and then was digested with the restriction enzyme Mnl I or BssHII. Eight crude lysates of urine and blood, which were proved hCMV positive by shell vial assay and were reconfirmed by PCR with primers
form the
pp65 coding region of the CMV genome. Were analyzed. Five different PCR-RFLP profiles were observed; two clinical isolates not amplified by alpha sequence primers, two clinical isolates identical with those of obtained by AD169, and three other
distinct
profiles. In conclusion, we showed the possibility of PCR-RFLP typing method for the differentiation of hCMV strain
